Solution

In a biochemical pathway each intermediate is the product of the previous step and is the substrate for the next step. So through biochemical pathway test we can analyze the mutated strain for a particular compound by comparing it with the wild type strain.

2.In this experiment the strains P, Q, R and S are mutant for methionine synthesis. Supplementing minimal media with an intermediate beyond step blocked in methionine auxotroph will allow it to grow. If mutant strain is blocked at early step more intermediate compound will allow the growth of strain and if the mutant strain is blocked at late stage fewer intermediate will allow the growth of the strain.

a) For strain P step iv is blocked

b) For strain Q step ii is blocked.

c) For strain R step v is blocked

c) For strain S step ii is blocked.

3.Each strain has a single mutation which is given as follows

a) strain P-met A+, met B+, met C- met E+.

b) strain Q met A+met B- met C+ met E+.

c) strain R met A+ met B+ met C+ met E-.

d) strain S met A+ met B- met C+ met E+.

4) Since Q and S both required o-succinyl-homoserine as supplement for growth the plating may give additional in formation about there growth pattern.

5)For strain R the step iv is blocked so its unable to grow at plate 5 in the absence of methionine as a supplement.
