What might a laboratory consider when determining an analytical threshold in RFU? Should this threshold be standard for all cases or vary according to the appearance of the raw data? What is stutter?

Solution

First of all, the amoount of DNA that is being analyzed is important as a low level of DNA could result in difficulty in separating a true level RFU peak from aberrations such as signal noise, etc. As establishment of minimum RFU varies from one laboratory to another, there is no as such standard threshold. The Stutter is referred to as the peaks in profile and the stutter values are derived from the homozygotes and the heterozygotes where alleles are separated by a minimum of 2 repeat units.
