8.) What kind of substances could interfere with the Dische’s test for DNA? How are these substances removed?
Solution
Dische Test is a test to determine the presence of DNA in a sample through spectrophometric analysis at A260/A280 absorbance ratio. It was found that the substances such as carbohydrates and proteins would interfere the test. It could be overcome by some biochemical methods such as a, colour reaction with tryptophan; b, standarized cysteine reaction as given in the original dische test and c, the micromethod using indole with DNA .
